Hypoxia-Inducible Factor-2α as a Novel Target in Renal Cell Carcinoma

Hypoxia-inducible factor (HIF), an important mediator of hypoxia response, is implicated in tumorigenesis the setting pseudohypoxia, such as inactivation von Hippel–Lindau tumor suppressor protein (pVHL), leading to development and progression clear cell renal carcinoma (ccRCC). Targeting downstream molecules HIF pathway, vascular endothelial growth (VEGF), has led improvement clinical outcome for patients with advanced ccRCC, but therapy thus far been limited by eventual resistance treatment failure. Following discovery HIF-2α playing a key role ccRCC carcinogenesis, inhibitors targeting have developed demonstrated encouraging efficacy safety profile trials. This review discusses promising therapeutic target ccRCC.
